Development of an Accurate Polymerase Chain Reaction (PCR) Assay for Genetic Sex Identification in Lumpfish (Cyclopterus lumpus) Based on Male-Specific Anti-Mullerian Hormone (amh) Gene

The production of lumpfish (Cyclopterus lumpus) has become crucial in controlling sea lice levels salmonid aquaculture. To improve their breeding, there is a need for early sex identification. genomic region containing the anti-Müllerian hormone (amh) gene was suggested as candidate master sex-determining lumpfish. However, genome contains three copies amh with ambiguous specificity, designated amh1, amh2, and amh3. study aims to analyse male-specific between these paralogues its application marker. In this study, we utilised polymerase chain reaction (PCR)-based assays identify markers estimate length genome. Our results indicate that specific approximately 27 kilobases (kb), encompassing amh1 amh2 genes, exhibits male whereas amh3 present both sexes. developed PCR-based genetic identification targeting exhibited over 97% concordance phenotypic records. Further experiments other members Cyclopteridae: Aptocyclus ventricosus, Eumicrotremus taranetzi, E. asperrimus revealed only A. ventricosus. Phylogenetic analyses using available Cyclopteridae sequences suggest arose lineage. findings, along development PCR test, hold great promise field aquaculture will also contribute significantly future investigations aiming enhance our understanding sex-determination system evolution chromosomes teleostean fish.
